As much as I hate to admit it, I'm absolutely clueless as to how to operate Ventrillo.

How do I assign and input content for Hotkeys? How do I use them properly?

I'm that clueless.

Illustrated guide for bonus points.

Its in the guide on that site.
When you go into the option all you do is push down the button you want for the hotkey, then click save

I think I got it, a short while I attempted to use Read A Book.

If no-one can hear you, try going into Setup -> Hardware Input Mixer and change the "Line" option to Line in (or, if it's already on Line in, change it to Stereo mix)

Towards the end I could here you, but it was almost to quiet to make out. So its something else you need to adjust now.
